Diversification of West Sumatera Sago Germplasm Base on Moleculer Characterization

ABSTRACT

Information about the genetic diversity of germplasm is needed to support the breeding program. Marker random amplified polymorphic DNA (RAPD) to improve efficiency in the early stages of selection and can shorten the time needed for breeding programs sago. This study aimed to obtain information on genetic diversity of sago in West Sumatra through RAPD technique. Genetic observations carried out by using 23 samples of DNA from various locations in West Sumatra, which were analyzed by using 4 primers selected. Making characters dendogram genotype was performed with the program NTSYS pc version 2.10. The results showed that the clustering analysis at 0.72 similarity level produced five major groups namely: the first group formed by the genotype of sago plants originating from Dharmasraya (DM1, DM2, DM3), Padang Pariaman (PP1, PP2, PP3), West Pasaman ( PB2, PB3), and East Pasaman (PT2); group formed by the two sago plant genotypes from areas east Pasaman (PT3), South Coast (PS1, PS2, PS4), Padang (P3), Agam (A1, A2 ); The third group is formed by the genotype of sago plants originating from the West Pasaman (PB1), and East Pasaman (PT1) The fourth group is formed by the genotype of sago plants originating from Agam (A3) and Mentawai (M), and the fifth group is formed by sago plant genotypes originating from the South Coast (PS3) and Padang (P1, P2).
